58 patients who were diagnosed as pancreatic cancer and advised to undergo surgery in the first peo-ple’s hospital of Yunnan province from January 2011 to June 2014 are enrolled.Tumor tissues are excised as ex-perimental specimens,and tissues adjacent to caner are set as control.Immunohistochemistry and Western blot are used to determine the expression of nerve growth factor(NGF)and precursor of nerve growth factor(ProNGF) in tissue of advanced pancreatic cancer.Gel -Pro analyzer 4 image analysis system is used to analyze the density of band.All the tests are repeated three times and then analyzed with the three IOD data by statistics.Immuno-histochemical and Western blot results show that NGF level in tumor tissues is higher than that in tissues adjacent to caner.In contrary,proNGF level in tumor tissues is lower than that in tissues adjacent to caner.Semiquantita-tive Western blot show that IOD of NGF level in tissues adjacent to caner and tumor tissues are 113.33 ±8.54 and 285.00 ±15.13,respectively.Compared with this,IOD of proNGF level are 245.00 ±7.55 和 86.33 ± 7.37,respectively.All the differences are statistically significant (P <0.05).Therefore,NGF /proNGF ratio in tumor tissue of advanced pancreatic cancer has changed and NGF has increased,proNGF has decreased,which may be one of the mechanisms leading to tumor cell apoptosis reduction and growth,invasion and enhanced mi-gration in advanced pancreatic cancer.
